What do you mean unlock? If you mean how do you activate it, then the answer is that it's not an active ability. If you have at least one point in it, then it increases your basic attack damage by dealing magic damage that scales by a small amount of AP. Then, every time you hit an enemy champion, minion, or neutral monster with a basic attack, it gains a stack of "blight" which appears as a purple "blob" above their head for a short amount of time (the blight changes for different skins. For example, if you're playing as Arctic Ops Varus, the blight will become blue grenades). You can have up to three stacks of blight per target and multiple targets can have stacks of blight at the same time. While a target or targets have blight stacks above their head, Varus (and only Varus) can pop the stacks of blight by hitting the affected target with another ability of his (his Q, E, or, if it's an enemy champion, his Ultimate). All stacks of blight will pop and deal extra damage on top of what the ability itself will do.
